2025年11月15日，国际细胞生物学权威期刊Autophagy (IF 14.3) 发表了武汉大学周荣家教授与程汉华教授课题组的最新研究成果，论文题为《Mitochondrial NAD⁺-mediated mitophagy alleviates type I interferon response to the cytosolic mitochondrial DNA》，该研究揭示了线粒体NAD+调控线粒体自噬及维持细胞稳态的分子机制和功能。

✍ 线粒体烟酰胺腺嘌呤二核苷酸（NAD⁺）在能量代谢中发挥核心作用，但其在线粒体自噬及固有免疫中的功能与作用机制仍不清楚。本研究发现，线粒体NAD⁺缺乏会引发线粒体自噬功能异常与炎症反应。研究证实，线粒体NAD⁺转运蛋白SLC25A51 缺失导致的线粒体NAD⁺耗竭，损伤BNIP3介导的线粒体自噬。线粒体NAD⁺的缺失会抑制SIRT3介导的FOXO3去乙酰化作用，进而导致BNIP3的转录水平下调，并阻碍MAP1LC3B/LC3B的募集。值得注意的是，在氧化应激下，线粒体NAD+缺乏会促进线粒体DNA（mtDNA）从线粒体释放到细胞质中，进而通过激活CGAS-STING1信号通路，加剧胞质游离mtDNA诱导的Ⅰ型干扰素应答。本研究通过细胞系的基因操作揭示了线粒体NAD⁺、线粒体自噬与mtDNA诱导炎症反应三者间全新的分子调控关联，表明线粒体NAD⁺有望成为缓解胞质游离mtDNA所引发无菌性炎症的潜在治疗靶点。因此，该研究为线粒体稳态、炎症和先天免疫之间的相互作用提供了新的见解。
